Strain Description
1.  The entire Septin12 genomic fragment in the bMQ251k06 BAC clone was obtained from the BACPAC Resource Center, and the fragment containing exon 2 to exon 10 was cloned into the pL253 targeting vector. A targeting vector with a serine-to-glutamate (S196E) substitution was introduced into exon 6 through site-directed mutagenesis, and loxP-neo-loxP cassette was inserted between exon 7 and exon 8 of Sept12. The target construct was electroporated into mouse embryonic stem cells to replace the wild-type allele ofSept12. Cell clones with the targeted allele were transfected with the cre-recombinase-expressing plasmid to remove the loxP-flanked neo cassette.

The homozygous knock-in mice displayed poor male fertility due to deformed sperm with defective motility and loss of annulus, a septin-based ring structure. It is suggested thatSept12 phosphorylation inhibits septin ring assembly during annulus biogenesis.
