Strain Description
1. A targeting vector was generated in which exons 2 and 3 of the Hspb7 gene wereloxp-flanked and a FLAG-tag sequence inserted before the stop codon of the Hspb7 gene. The construct was electroporated into 129S6/SvEvTac -derived TC1 embryonic stem (ES) cells.Blastocyst injection of ES cells carried targeted allele was used to obtain chimera. The chimeric males were backcrossed to C57BL/6 females. The backcrosses involved mice constitutively expressing the FLP recombinase to remove the neomycin cassette from targeted allele.
2. The offspring of muscle specific-Hspb7 gene knockout began to die at the age of postnatal 1 week. The survival curve showed a wide spectrum of lethality in muscle specific-Hspb7 gene knockout mice as shown in reference.
