Strain Description
1. 
This reporter mouse may serve as an early detection of drug- and food-induced nephrotoxicity and as a mouse model to investigate tubular cell injury, acute kidney injury (AKI) and metabolism.
2.  The BAC-base transgene was designed NanoLuc driven by Miox genes. Miox was selected as the host promoter for renal-specific transgene expression. NanoLuc is inserted into exon 1 of Miox gene and followed after ATG start codon. The transgene was microinjected into C57BL/6J embryos. The founder was crossed with C57BL/6J for germline transmission. The expression levels ofMiox in serum, urine, and kidney were no significant difference between C57BL/6J and heterozygote.
3.  .A strong luminescence intensity was observed in kidney localized in the renal tubule cells, but not in wild type. NanoLuciferase is specifically expressed in the serum, urine, and kidney tissue of this strain.
4.  Luminescence level in blood was significantly increased after 1 day of cisplatin treatment and 3 days of Aristolochic acid (AAI) treatment. The sensitivity is earlier than the test on creatinine and BUN. It can not only reflect nephrotoxicity, but also can be detected before the pathological change of renal tubules damage.
5.  Luminescence produced from Miox-NanoLuc had good stability at room temperature, 4∘C, 20∘C and repeated freeze-thaw cycles.
