Strain Description
1.  A targeting vector containing aloxP-flanked neomycin-resistant (Neo) cassette inserted the intron 18 of Scube1 and an additional loxP site in the upstream of intron 14 of the gene. Successful targeting of embryonic stem cells with vector above mentioned resulted in a conditional allele including the Neo cassette. Additional Cre-mediated recombination in embryonic stem cells deleted the Neo cassette and produce a conditional allele (Scuble1Flox) that induce 2 loxP sites flanking exons 15 to 18 which encode a part of the spacer region and the complete 3 cysteine rich (CR) repeat motifs of Scube1. Heterozygous (Scub1/+) mice were generated by crossing male protamine (Prm)-cre; Scuble1Flox/+ mice to wild-type female mice.
2.  The coding sequence for portion of the spacer region and the 3 CR repeats (exons 15-18) were successfully deleted, yet the COOH-terminal CUB (complement protein C1r/C1s, Uegf, and Bmp1) domain was transcribed via in-frame coding in mutant (△/△) mice platelets. The plasma concentration of SCUBE1 protein was around 150ng/mL in wild-type (+/+), whereas soluble SCUBE1 was undetectable in mutant mice (△/△), and both wild-type and truncated protein could be released from α-granules and tether on the platelet surface upon activation.
3.  This strain showed normal development, survival, and reproduction and no spontaneous bleeding or thrombosis. 
4.  Blood cell counts (including platelets) and hematologic and coagulation parameters, as well as protein or mRNA levels of major platelet surface receptors, including glycoprotein VI, glycoprotein IX, glycoprotein Ia (α2), β3 integrin, and ADP receptors (P2Y1 and P2Y12), were similar between wild-type (+/+) and mutant mice (△/△). In addition, platelet aggregation was indistinguishable in platelet-rich plasma (PRR) samples from wild-type (+/+) and mutant mice (△/△). Furthermore, electron microscopy revealed no apparent differences in ultrastructure or the numbers ofα- or dense granules between wild-type (+/+) and mutant mice (△/△).
